What is the PCR COVID-19 Test?

PCR stands for a polymerase chain reaction and is the gold-standard method for detecting the COVID-19 virus from any sample (nasal swab, throat swab, or saliva sample). This type of molecular testing identifies the DNA or RNA to determine if the virus is active in the patient. PCR tests can find the virus in the early stages, which can help prevent the spread of COVID-19.

How Does PCR COVID-19 Work?

Currently, patients may receive free testing at health centers throughout the United States. The healthcare provider takes a swab of the nasal passage. The SARS-CoV-2 virus uses RNA, which the lab converts from single-stranded RNA to double-stranded DNA using reverse transcription. The lab then separates the two DNA template strands. Primers then attach to the DNA strands.

If the primers match the DNA, the DNA amplifies, and the sample is positive for COVID-19. If the sample shows no amplification, the result is negative for COVID-19. The primer can only amplify from viral material, making false positives extremely rare.

What Are Inconclusive and False Negative Results?

Due to the highly sensitive nature of the COVID-19 testing technology, there are rare cases where your results may not immediately produce a conclusive result.

When a result is "inconclusive," we recommend repeating COVID-19 testing. The test may have picked up the residual virus from someone recovering from a COVID-19 infection and who is, in fact, no longer infected. Alternatively, the test could have picked up a minute amount of contaminant during the collection or processing.

"False-negative" results can occur when a person has COVID-19, but the test results are negative. At the very early stages of an infection, the amount of virus can be too low for the COVID-19 test to detect the virus.

We recommend repeating the test if you develop symptoms, even if a recent COVID-19 test returned negative. Use a PCR test in this situation because antigen tests are more likely to give a "false negative" result.
